HumanNeuralStemCells(Cat.#1650)fromScienCellResearchLaboratoriesaredifferentiatedfromahumaninducedpluripotentstemcellline(hiPSC),whichisgeneratedusingmRNAreprogrammingtechnologyfromHumanDermalFibroblasts(HDF).ThemonolayerHDF-hiPSCareefficientlyconvertedtoneuralepitheliumusingHPSCNeuralInductionMedium(PSCNIM,Cat.#5931),aserum-freemediumforrapidandefficientneuralinductionofhumanpluripotentstemcells(hPSCs).Synergisticinhibitionofglycogensynthasekinase3(GSK3)andtransforminggrowthfactorb(TGF-b)differentiatesHDF-hiPSCstohomogenousneuralstemcells(NSC)inthepresenceofhumanleukemiainhibitoryfactor(LIF)within7days.ThederivedNSCarecharacterizedbyimmunofluorescencewithantibodiesspecifictonestinandSOX2.Thecellpopulationishighlypure:>90%ofcellsexpressNestinand>80%ofcellsareSOX2positive.Afterreviving,NSCcanbemaintainedinHPSCNeuralInductionMediumasanadherentcultureorformneuralspheresinEGFandbFGFcontainingneuralstemcellmedium.Thecellsareabletoformrosetteandneuraltube-likestructuresafterbeingpassagedandplatedathighdensityonMatrigelcoatedculturevesselsinneuralstemmediumcontainingEGFandbFGF.HiPSC-NSCcanbefurtherexpandedformonthsasamonolayeronMatrigelTMcoatedculturevesselsorinsuspensionasneuralsphereswhenseededonultra-lowbindingculturevesselsinneuralstemcellmedium.NSCaremultipotentandabletodifferentiateintovariousneuronalandglialsubtypes.Specificpatterningcues,suchasSHH,retinoicacidandFGF8,canbeaddedafterrevivingtodirectthecellstodifferentneurallineages.HiPSC-NSCarecryopreservedatpassage0anddeliveredfrozen.Eachvialcontains>1x10^6cellsin1mlvolume.CellsarenegativeforHIV-1,HBV,HCV,mycoplasma,bacteria,yeastandfungi.
