Glutathioneperoxidase(GPx)isanantioxidantenzymelocatedincytoplasmaandmitochondria.Itcatalyzesthereductionofhydrogenperoxide(H2O2)andavarietyoforganichydroperoxides(R-O-O-H)towaterandthecorrespondingstablealcohols(R-OH)usingglutathioneasthereducingreagent,thusinhibitingtheformationoffreerADIcalsandprotectingthecellsfromoxidativedamage.TheSciencellGPxAssayprovidesanindirectmethodtoquantifytheactivityoftotalglutathioneperoxidase.Inthisassay,oxidizedglutathione,produceduponreductionofperoxidesubstratebyglutathioneperoxidase,isthenrecycledbacktoglutathioneusingglutathionereductaseandNADPH.TheoxidationofNADPHtoNADP+ismonitoredbyadecreaseintheabsorbanceat340nm.Sinceallotherreagentsareprovidedinexcess,theGPxactivityisrate-limiting.TherateofdecreaseinA340nmisdirectlyproportionaltotheGPxactivity.Inthisassay,cumenehydroperoxideisusedastheperoxidesubstrate,sothetotalGPx(seleniumandnon-seleniumcontaining)activityinavarietyofsamplescanbemeasured.
