Malatedehydrogenase(MDH2)catalyzesthelastreactionoftheTCAorKrebscycle,convertingmalateandNAD+tooxaloacetateandNADHinthemitochondria.Inreversingthereaction,MDHisalsoinvolvedingluconeogenesis,allowingoxaloacetate/malatetoleavethemitochondria.Onceinthecytosol,themalateisoxidizedbacktooxaloacetatebycytosolicMDHorMDH1,whichisfollowedbyconversionofoxaloacetatetophosphoenolpyruvatebyphosphoenolpyruvatecarboxykinase(PEPCK).Theinterconversionbetweenmalateandoxaloacetatealsoconstitutestheessentialstepsofthemalate-aspartateshuttle.Thiscolorimetricassayisbasedonmalatedehydrogenase-catalyzedoxidationofmalate,wheretheresultingNADHcanthenconvertanearlycolorlessprobetoacoloredproduct;theintensityofthecoloredproductisproportionaltotheamountofMDHinthesample,exhibitingmaximumabsorbanceat440nm.
